چکیده

The purpose of the research is to identify perinatal infection in dynamics, and assess number genetic status bovine leukemia proviruses isolated from young animals, correlations between some indicators infectious process based on gene diagnostics methods. Materials methods . We used material cattle different age groups: 1, calves (30–40 minutes after birth before colostrum 15 45 days); 2, heifers (not older than two years). Radial immunodiffusion (RID), real-time polymerase chain reaction (PCR), phylogenetic analysis were used. Results discussion An assessment given for cattle. detection rate was 4.15% (PCR) 1.09% (RID); 1.1% 0.88% (RID) heifers. A 36-fold decrease found positive dynamics (2013–2022) 14.5 0.4% with passing through 0% (2020) being at level (2022). proviral load ranged 2.02 × 104 8.38 106 GE/mL blood examined animals. BLV isolates obtained shown belong genotypes, GIV GVII (env), clade 1 (pol). assessed an overestimation by a factor three animals under years (3.83 106GE/mL) relative that 1-month-old (1.3 GE/mL), nine It important develop algorithms increase effectiveness routine tools prevent spread this retrovirus early stage, which confirmed detected over time. provirus higher calves; multiparous dairy cows nulliparous quantitative animals’ genotype those variant BLV.

Estimation of bovine leukemia virus (BLV) proviral load harbored by lymphocyte subpopulations in BLV-infected cattle at the subclinical stage of enzootic bovine leucosis using BLV-CoCoMo-qPCR

BACKGROUND Bovine leukemia virus (BLV) is associated with enzootic bovine leukosis (EBL), which is the most common neoplastic disease of cattle. BLV infection may remain clinically silent at the aleukemic (AL) stage, cause persistent lymphocytosis (PL), or, more rarely, B cell lymphoma. BLV has been identified in B cells, CD2+ T cells, CD3+ T cells, CD4+ T cells, CD8+ T cells, γ/δ T cells, mono...

Blood dendritic cells in cattle infected with bovine leukemia virus (BLV): isolation and phenotyping.

Dendritic cells (DCs) are most potent antigen presenting cells (APCs) with unique ability to prime effective immune responses. They express higher levels of MHC class II and accesory molecules on their surface, than other professional APCs. The investigations were performed on DCs generated from blood with the use of microbeads magnetically labeled with mouse anti human CD14. Flow cytometry was...

Comparison of bovine leukemia virus (BLV) and CMV promoter-driven reporter gene expression in BLV-infected and non-infected cells

BACKGROUND: Viral promoters are used in mammalian expression vectors because they generally have strong activity in a wide variety of cells of differing tissues and species. METHODS: The utility of the BLV LTR/promoter (BLVp) for use in mammalian expression vectors was investigated through direct comparison to the CMV promoter (CMVp). Promoter activity was measured using luciferase assays of ce...

Genetic variation of bovine leukemia virus (BLV) after replication in cell culture and experimental animals.

This article reports the selection of bovine leukemia virus (BLV) variants after continuous passage in cell lines or experimental animals. Two wild BLV strains isolated from 2 naturally infected Holstein dairy cows in Brazil (cow codes: 485 and 141) were used for the experimental infection of 1 sheep and FLK cells, and 1 rabbit and CC81 cells. Viral DNA was isolated several months after infecti...

MicroRNAs Encoded by Bovine Leukemia Virus (BLV) Are Associated with Reduced Expression of B Cell Transcriptional Regulators in Dairy Cattle Naturally Infected with BLV

Bovine leukemia virus (BLV) is estimated to infect over 83% of dairy herds and over 40% of all dairy cows in the United States. While, BLV only causes leukemia in a small proportion of animals, research indicates that BLV+ cattle exhibit reduced milk production and longevity that is distinct from lymphoma development. It is hypothesized that BLV negatively affects production by interfering with...
